目的 为脑膜瘤X 刀的个体化治疗寻找最佳量 效关系和最佳时 效关系。方法 8例病人的脑膜瘤细胞进行原代培养。将培养的脑膜瘤细胞分为阴性对照组、5Gy、10Gy、15Gy、2 0Gy、2 5Gy组和 30Gy组 ,培养 7~ 9天后采用单次X线照射 ,于照射后 2h、6h、12h、2 4h、48h和 72h采用FCM检测细胞凋亡和细胞存活情况 ,采用免疫组化(ABC)法测试PCNA、BcL 2和p5 3基因的表达。结果 发现原代培养的脑膜瘤细胞于培养后的第 7~ 2 1天 ,细胞存活率可达 80 %~ 92 %。X线诱导细胞凋亡平均最佳剂量为 2 0Gy,平均最佳检测时间为 2 4h ,平均凋亡率为 5 7 6 5 %± 15 5 2 %。结论 不同的个体需要的细胞凋亡的X线剂量有很大差异。尽管影响剂量选择的因素很多 ,但本研究的结果显示可能与一些基因的表达有一定关系
Objective To find the best dose-effect relationship and the best time-effect relationship for the individualized treatment of meningioma X knife. Methods Eight patients with meningioma were cultured in primary culture. The cultured meningioma cells were divided into negative control group, 5Gy, 10Gy, 15Gy, 20Gy, 25Gy and 30Gy groups. After cultured for 7-9 days, Cell apoptosis and cell survival were detected by FCM at 4h, 48h and 72h. The expression of PCNA, BcL 2 and p5 3 genes were detected by immunohistochemistry (ABC). The results showed that the survival rate of primary meningioma cells reached 80% ~ 92% on the 7th ~ 21st day after culture. The average best dose of X-ray induced apoptosis was 20 Gy, the average best detection time was 24 h and the average apoptosis rate was 5765% ± 1552%. Conclusion Different individuals may have very different doses of X-ray for apoptosis. Although there are many factors that influence the choice of dose, the results of this study may have some relation to the expression of some genes
