目的 比较DC对诱导4 d(L4) 和诱导7 d(L7) 的LPAK 细胞体外杀伤人肝癌细胞株BEL- 7402(B) 的杀伤力和杀伤模式的影响.方法 L4 组为B+ L4 ,LD4 组为L4 组+ DC;L7 组为B+ L7 ,LD7 组为L7 组+ DC.L4 和L7 与B的效靶比均为5∶1 和10∶1两种. 采用杀伤细胞检测技术及电镜技术,比较各组的杀伤效应和杀伤模式.结果 各组的细胞毒活性为LD4 > L4( P< 0-01) ,LD7 > L7( P< 0-01) .L4 组和LD4 组的BEL7402 细胞呈现不同程度的变性坏死,L7 组和LD7 组的BEL7402 细胞则呈现不同程度的凋亡改变.结论 DC对不同诱导时间的LPAK 细胞体外杀伤肿瘤细胞都有明显的促进作用,但并不改变LPAK 细胞对肿瘤细胞的杀伤模式.
Objective To compare the effects of DC on the killing and killing of human hepatocellular carcinoma cell line BEL-7402 (B) induced by LPAK cells in vitro for 4 days (L 4) and 7 days (L 7). Methods L4 group was B+L-4, LD4 group was L4 group+DC, L7 group was B+L-7, and LD7 group was L7 group+DC. The ratio of L 4 to L 7 to B was 5:1 and 10:1. Using killer cell detection technology and electron microscopy technology, the killing effect and killing pattern of each group were compared. Results The cytotoxic activity of each group was LD4 > L4 (P<0-01), and LD7 > L7 (P<0-01). The BEL-7402 cells in L4 and LD4 groups showed degeneration and necrosis in different degrees. The BEL-7402 cells in L7 and LD7 groups showed varying degrees of apoptosis. Conclusion DCs can obviously promote the killing of tumor cells in vitro by LPAK cells with different induction time, but they do not change the killing mode of tumor cells by LPAK cells.
