目的:探讨益气活血复方愈心痛胶囊对缺血心肌组织内皮素转换酶(ECE)、一氧化氮合酶(NOS)基因表达和铜锌超氧化物歧化酶(铜锌-SOD)免疫组织化学反应的影响,阐明愈心痛胶囊抗心肌缺血的作用机制。方法:采用结扎麻醉犬冠状动脉左前降支中段的方式,造成实验性犬急性心肌缺血病理模型,以原位杂交技术检测缺血心肌中ECE和NOSmRNA的表达,用免疫组织化学方法观察缺血心肌中铜锌-SOD阳性反应程度。结果:在缺血心肌组织中,ECEmRNA呈高表达,NOSmRNA呈低表达,愈心痛可提高缺血心肌中NOSmRNA表达水平,削弱ECEmRNA的异常表达,从而保护缺血心肌;同时,缺血心肌中铜锌-SOD免疫反应减弱,而愈心痛可显著增强缺血心肌组织中铜锌-SOD阳性反应程度,提高SOD活性。结论:愈心痛胶囊的上述抗心肌缺血的作用,部分说明了益气活血方药的作用机理,提示该方可能对缺血性心脏病的治疗具有潜在的临床应用价值。
Objective: To investigate the expression of endothelin-converting enzyme (ECE) and nitric oxide synthase (NOS) genes and copper-zinc superoxide dismutase (Cu-Zn-SOD) immunohistochemistry in Yiqi Huoxue Fufang Yuxintong Capsules for myocardial ischemia. The effect of the reaction clarifies the mechanism of action of Yuxintong capsule against myocardial ischemia. METHODS: The pathological model of acute myocardial ischemia in experimental dogs was induced by ligation of the middle segment of the left anterior descending coronary artery in canine dogs. The expression of ECE and NOS mRNA in ischemic myocardium was detected by in situ hybridization. Immunohistochemistry was used to observe ischemia. The degree of copper-zinc-SOD positive reaction in myocardium. RESULTS: In ischemic myocardium, ECE mRNA was highly expressed and NOSmRNA was lowly expressed. Yuxintong can increase the expression of NOSmRNA in ischemic myocardium and weaken the abnormal expression of ECE mRNA, thus protecting ischemic myocardium; meanwhile, copper in ischemic myocardium The zinc-SOD immune response was weakened, and Yuxintong significantly increased the level of copper-zinc-SOD positive reaction in ischemic myocardium and increased SOD activity. Conclusion: The above anti-myocardial ischemia effect of Yuxintong capsule partially explained the mechanism of Yiqihuoxue prescription, suggesting that this party may have potential clinical application value in the treatment of ischemic heart disease.
