目的 研究光性屈光性角膜切削术(PRK)后,转移生长因子β受体Ⅱ(TGFβRⅡ) 和表皮生长因子受体(EGFR)mRNA 在角膜的表达变化,探讨角膜上皮下混浊(haze) 形成的发生机制。方法 新西兰白兔16 只,随机分成4 组。其中12 只施行PRK,分别于术后1 、2、3 个月用裂隙灯显微镜观察haze 形成情况,并用原位核酸分子杂交方法,检测角膜上皮和基质TGFβRⅡ和EGFRmRNA的表达。结果 PRK后1 个月已有haze 形成,2 个月时haze 最明显,术后3 个月haze 减轻或消失。正常兔角膜上皮和基质有TGFβRⅡ和EGFRmRNA表达,PRK后角膜TGFβRⅡ和EGFRmRNA表达增加,且以术后1、2 个月表达最明显。TGFβRⅡ和EGFRmRNA表达强弱与形成haze 的轻重相关。结论 TGFβRⅡ和EGFR参与PRK后伤口愈合过程,且调节着haze 的形成和发展
Objective To study the changes of corneal expression of TGFβRⅡ and epidermal growth factor receptor (EGFR) mRNA in cornea after photorefractive keratectomy (PRK) ) Formed by the mechanism of occurrence. Methods Sixteen New Zealand white rabbits were randomly divided into four groups. PRK was performed in 12 of them, haze formation was observed with slit lamp microscopy at 1, 2 and 3 months after operation, and the expression of TGF-βRII and EGFR mRNA in corneal epithelium and stroma was detected by in situ hybridization. Results One month after PRK, haze was formed, haze was the most obvious at 2 months, haze was reduced or disappeared 3 months after operation. Normal rabbits corneal epithelium and matrix have TGF β R Ⅱ and EGFR mRNA expression, PRK corneal TGF β R Ⅱ and EGFR mRNA expression increased, and the expression was most obvious after 1, 2 months. The expression of TGF β R Ⅱ and EGFR mRNA strength and the formation of haze severity related. Conclusion TGF β R Ⅱ and EGFR participate in wound healing process after PRK, and regulate the formation and development of haze
