目的:研究肿瘤干细胞标记物USP22 mRNA在不同级别膀胱移行细胞癌和正常膀胱黏膜中的表达差异,并探讨其与肿瘤分级,临床分期的关系。方法:分别抽提不同级别的膀胱移行细胞癌组织标本和正常膀胱黏膜组织标本的总RNA,经RT为cDNA后,采用qRT-PCR法检测USP22 mRNA的表达情况。结果:正常膀胱黏膜组织中USP22的表达明显低于膀胱肿瘤组织,几乎不表达;不同级别膀胱肿瘤组织中,低度恶性潜能的乳头状尿路上皮癌和低级尿路上皮癌中USP22的表达要明显高于其在高级尿路上皮癌中的表达;特别是Ⅲ级膀胱移行细胞癌组,USP22的表达仅略高于其在正常膀胱黏膜中的表达。结论:USP22可能是正常干细胞向肿瘤干细胞恶变的关键调节分子,可能成为一个新的诊断移行上皮癌侵袭性的有效指标。干扰其蛋白质合成将成为一个抗肿瘤生长的新靶点。
OBJECTIVE: To study the expression differences of tumor stem cell marker USP22 mRNA in different grades of bladder transitional cell carcinoma and normal bladder mucosa, and to explore its relationship with tumor grade and clinical stage. Methods: Total RNA was extracted from bladder transitional cell carcinoma specimens and normal bladder mucosa specimens at different levels respectively. After RT cDNAs, the expression of USP22 mRNA was detected by qRT-PCR. Results: The expression of USP22 in normal bladder mucosa tissues was significantly lower than that in bladder tumor tissues and was almost not expressed. The expression of USP22 in low grade malignant papillary urothelial carcinoma and low grade urothelial carcinoma in different grades of bladder tumor tissues Was significantly higher than its expression in advanced urothelial carcinoma; especially in grade Ⅲ bladder transitional cell carcinoma, the expression of USP22 was only slightly higher than that in normal bladder mucosa. CONCLUSION: USP22 may be a key regulator of normal stem cell malignant transformation of tumor stem cells, which may be a new effective indicator to diagnose the invasiveness of transitional cell carcinoma. Interfere with its protein synthesis will be a new target for anti-tumor growth.
