为改善肺癌化疗效果,用FCM技术分析研究胃肠外营养(PN)对人肺腺癌裸鼠模型LAX-83细胞动力学的影响。腹腔注射PN 1ml,每8小时一次,共72小时,分别于投药后24、48和72小时取佯分析细胞周期各时相细胞百分比。对照组和投药后24、48、72小时DNA合成期细胞百分比(S%)分别为43.1±3.2%、53.4±10.2%、36.4±4.8%和56.6±9.7%。S%随时间呈波浪式变化,而且48小时和72小时的S%与对照组之间有显著性差异(P<0.05)。结果提示PN可影响肺癌细胞动力学,为改善肺癌细胞对周期特异性化疗药物反应性提供契机。此外,S%呈波浪式变化根示在PN刺激下,细胞周期中的部分细胞被同步化。
In order to improve the effect of chemotherapy in lung cancer, the effect of parenteral nutrition (PN) on the cell kinetics of human lung adenocarcinoma model LAX-83 was analyzed by FCM. Intraperitoneal injection of PN 1ml, once every 8 hours, for a total of 72 hours, were taken at 24, 48 and 72 hours after administration to analyze the percentage of cells in each phase of the cell cycle. The percentage of cells in the DNA synthesis phase (S%) in the control group and at 24, 48, and 72 hours after administration were 43.1±3.2%, 53.4±10.2%, 36.4±4.8%, and 56.6±9.7%, respectively. The S% changes wave-like over time, and there is a significant difference between the S% at 48 hours and 72 hours and the control group (P<0.05). The results suggest that PN can affect the cell kinetics of lung cancer, and provide an opportunity to improve the response of lung cancer cells to cycle-specific chemotherapy drugs. In addition, the wave-like change of S% is rooted in PN stimulation, and some cells in the cell cycle are synchronized.
