目的:探讨替米沙坦对糖尿病心肌病(DCM)大鼠心脏的保护作用。方法:30只健康雄性Wistar大鼠,随机分成空白组(n=10)和糖尿病组(n=20),糖尿病组用链脲佐菌素(STZ)腹腔注射建模,成功后随机分成对照组(n=10)及治疗组(n=10)。治疗组用替米沙坦(20mg/kg)每天灌胃治疗一次,空白组和对照组给予相同体积的生理盐水灌胃,给药16周后彩色多普勒检测心功能,光镜下观察心肌病理改变,电镜下观察心肌超微结构,原位缺口末端标记法(TUNEL法)检测心肌细胞凋亡,免疫组化法检测心肌组织转化生长因子β1(TGF-β1)的表达。结果:糖尿病对照组心功能明显下降,心肌出现明显病理学改变,凋亡细胞增多(P<0.05),TGF-β1的表达增加(P<0.05);而替米沙坦治疗组,心功能无明显改变,心肌病理改变较对照组明显减轻,凋亡细胞减少(P<0.05),TGF-β1的表达减少(P<0.05)。结论:替米沙坦对糖尿病心肌病大鼠心肌有保护作用,改善心功能。
Objective: To investigate the protective effect of telmisartan on diabetic cardiomyopathy (DCM) rats. Methods: Thirty healthy male Wistar rats were randomly divided into blank group (n = 10) and diabetic group (n = 20). The diabetic rats were injected intraperitoneally with streptozotocin (STZ) (n = 10) and treatment group (n = 10). The treatment group was treated with telmisartan (20mg / kg) once daily by gavage. The blank group and the control group were given the same volume of physiological saline for gavage. After 16 weeks’ administration, the color Doppler was used to detect the cardiac function. The ultrastructures of myocardium were observed under electron microscope. The apoptosis of myocardial cells was detected by TUNEL method. The expression of transforming growth factor-β1 (TGF-β1) in myocardium was detected by immunohistochemistry. Results: The cardiac function of diabetic control group was significantly decreased, the pathological changes of myocardium were obvious, the number of apoptotic cells was increased (P <0.05) and the expression of TGF-β1 was increased (P <0.05) (P <0.05), while the expression of TGF-β1 decreased (P <0.05). Conclusion: Telmisartan protects the myocardium of diabetic cardiomyopathy rats and improves cardiac function.
