本实验用胰蛋白酶和胶原酶分离得到大鼠睾丸巨噬细胞(MФ)和 Leydig 细胞,用腹腔冲洗法获得大鼠腹腔 MФ,用 McCoy 5 A 培养液进行培养。结果显示,当睾丸 MФ和 Leydig 细胞共同培养时,Leydig 细胞的睾丸酮分泌量为9.1±0.9 pmoI/10~0细胞,(?)对照组(6.3±1.5 pmol/10~0细胞)显著增加。相反,当腹腔 MФ和 Leydig 细胞共同培养时,L dig 细胞的睾丸酮分泌量为4.1±0.3 pmol/10~0细胞,比对照组(14.2±0.4 pmol/10~0细胞)显著下降。结果表明,大鼠睾丸MΦ在体外培养时能促进 Leydig 细胞分泌睾丸酮,而腹腔 MΦ则抑制 Leydig 细胞分泌睾丸酮.本文的研究对不同部位的巨噬细胞具有异质性提供了又一例证。
In this experiment, rat testicular macrophages (MФ) and Leydig cells were isolated by trypsin and collagenase. The peritoneal MФ was obtained by peritoneal washing and cultured in McCoy 5 A medium. The results showed that when testicular MΦ and Leydig cells were co-cultured, the amount of testosterone secretion in Leydig cells was 9.1 ± 0.9 pmo I / 10-0 cells (6.3 ± 1.5 pmol / 10-0 cells). In contrast, the amount of testosterone secretion in L dig cells was significantly decreased when co-cultured with Mφ and Leydig cells (4.1 ± 0.3 pmol / 10-0 cells), compared with the control group (14.2 ± 0.4 pmol / 10-0 cells). The results showed that rat testicular MΦ can promote the secretion of testosterone in Leydig cells while MΦ in the peritoneal cavity inhibits the secretion of testosterone in Leydig cells.This study provides another example of heterogeneity of macrophages in different parts.
