目的 :探讨转化生长因子β1 (TGFβ1 )及p2 7kip1 在胃癌癌变过程中的作用。方法 :采用逆转录PCR技术分别检测胃粘膜正常组织、单纯性增生、不典型增生及胃癌组织标本中TGFβ1 和p2 7kip1 mRNA表达水平。结果 :正常胃粘膜、单纯性增生、不典型增生及胃癌组织均有TGFβ1 和p2 7kip1 mRNA表达 ,但TGFβ1 和p2 7kip1 的mRNA表达水平在上述 4种组织中均依次逐渐下降 ,胃癌组的表达水平最低 ,与其它三组比较差异有显著性 (P <0 .0 5 )。结论 :TGFβ1 和p2 7kip1 对胃癌的发生发展可能起重要作用 ;用半定量PCR技术能有效地分析组织内mRNA表达水平
Objective: To investigate the role of transforming growth factor β1 (TGFβ1) and p27kip1 in the carcinogenesis of gastric cancer. Methods: Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of TGFβ1 and p27kip1 mRNA in gastric mucosa normal tissue, simple hyperplasia, atypical hyperplasia and gastric cancer tissue respectively. Results: The mRNA expressions of TGFβ1 and p27kip1 in normal gastric mucosa, simple hyperplasia, atypical hyperplasia and gastric cancer tissues were all decreased, but the mRNA levels of TGFβ1 and p2 7kip1 decreased gradually in the above four tissues, The lowest, with the other three groups the difference was significant (P <0. 05). Conclusion: TGFβ1 and p2 7kip1 may play an important role in the occurrence and development of gastric cancer. Semi-quantitative PCR can effectively analyze the mRNA expression level in tissues
