目的:探讨胶原三股螺旋重复蛋白1(collagen triple helix repeat containing 1,CTHRC1)在宫颈癌组织中表达情况及其临床意义。方法:宫颈癌患者39例(宫颈癌组),正常体检者(对照组)、慢性宫颈炎(宫颈炎组)和宫颈上皮内瘤样病变(CIN组)患者各30例,采用免疫组织化学和ELISA方法检测CTHRC1蛋白的表达情况,分析其表达与病理分级、临床分期及淋巴结转移的关系。结果:CTHRC1主要表达在细胞质,宫颈癌组36例(92.3%)呈阳性表达,CIN组18例(60%)阳性表达,宫颈炎组2例(6.67%)阳性表达,对照组无阳性表达,宫颈癌组和其他组比较差异均有统计学意义(P<0.05);宫颈癌组CTHRC1蛋白表达水平明显高于对照组、宫颈炎组及CIN组(P<0.05);CIN组、宫颈癌组CTHRC1蛋白表达水平呈上升趋势(P<0.05)。CTHRC1在宫颈癌组织中表达在病理分级、临床分型方面差异有统计学意义(P<0.05),在有无淋巴结转移方面差异无统计学意义(P>0.05)。结论:CTHRC1表达水平与宫颈癌的病理分级及临床分期有关,有望成为宫颈癌发生、发展的重要指标之一。
Objective: To investigate the expression of collagen triple helix repeat containing 1 (CTHRC1) in cervical cancer and its clinical significance. Methods: Thirty nine patients (cervical cancer group), 30 normal subjects (control group), 30 chronic cervicitis patients (cervicitis group) and 30 patients with cervical intraepithelial neoplasia (CIN group) were enrolled in this study. Immunohistochemistry The expression of CTHRC1 protein was detected by ELISA. The relationship between the expression of CTHRC1 protein and pathological grade, clinical stage and lymph node metastasis was analyzed. Results: CTHRC1 was mainly expressed in the cytoplasm. The expression of CTHRC1 was positive in 36 cases (92.3%) of cervical cancer group, 18 cases (60%) in CIN group, 2 cases (6.67%) in cervicitis group, (P <0.05). The expression of CTHRC1 protein in cervical cancer group was significantly higher than that in control group, cervicitis group and CIN group (P <0.05), while CIN group and cervical cancer group CTHRC1 protein expression levels showed an upward trend (P <0.05). The expression of CTHRC1 in cervical cancer was significantly different in pathological grade and clinical classification (P <0.05), but there was no significant difference in the presence or absence of lymph node metastasis (P> 0.05). Conclusion: The expression of CTHRC1 is related to the pathological grade and clinical stage of cervical cancer, which is expected to be one of the important indicators for the occurrence and development of cervical cancer.
