目的研究四氢呋喃(tetrahydrofuran,THF)与牛血清白蛋白(bovine serum albumin,BSA)相互作用的机制,探讨温度对其反应的影响。方法移取的BSA于石英比色皿中,搅拌混合均匀,避光静置,分别在25℃(298 K)、29℃(302 K)、33℃(306 K)、37℃(310 K)下以280 nm为激发波长,在LS-55荧光分光光度计上扫描280~500 nm波长范围内的发射光谱。结果不同温度下的结合猝灭常数(Ksv)为(3.466~6.317)×104L/mol,猝灭常数随温度的升高而减小。结论四氢呋喃对BSA的猝灭作用是属于静态猝灭。
Objective To study the mechanism of the interaction between tetrahydrofuran (THF) and bovine serum albumin (BSA) and to investigate the effect of temperature on the reaction. Methods The BSA was extracted from the quartz cuvette and mixed well at 25 ℃ (298 K), 29 ℃ (302 K), 33 ℃ (306 K) and 37 ℃ (310 K) Under 280 nm excitation wavelength, LS-55 fluorescence spectrophotometer scan the emission spectrum in the wavelength range of 280 ~ 500 nm. Results Under different temperatures, the quenching constant (Ksv) was (3.466 ~ 6.317) × 104 L / mol. The quenching constant decreased with increasing temperature. Conclusion The quenching effect of tetrahydrofuran on BSA belongs to static quenching.
