本实验以体外培养大鼠肝细胞硫代乙酰胺(TAA)损伤为模型,采用四甲基偶氮唑盐(MTT)显色法,观察比较了成年小型猪部分肝切除后,取正在再生肝脏制得的肝细胞生长因子(PRHGF)和市场销售的乳猪肝肝细胞生长因子(PHGF)对肝细胞损伤保护作用的大小。结果表明,PRHGF和PHGF对TAA引起的肝细胞损伤均有保护作用,但PRHGF的最大效应高于PHGF组,提示PRHGF是一种有效的抗TAA肝损伤和刺激损伤后肝细胞再生的药物。
In this study, rat hepatocyte thioacetamide (TAA) injury was cultured in vitro as a model, using MTT colorimetric method to observe and compare the partial liver resection of adult miniature pigs, Preparation of hepatocyte growth factor (PRHGF) and the commercially available suckling pig liver hepatocyte growth factor (PHGF) on the protective effect of liver cell damage. The results showed that PRHGF and PHGF had protective effects on TAA-induced hepatocyte injury, but the maximum effect of PRHGF was higher than that of PHGF group, suggesting PRHGF is an effective anti-TAA liver injury and stimulating hepatocyte regeneration after injury.
