目的:研究局部稳定表达OPG在种植体周围骨再生修复过程中的作用。方法:将66只SD大鼠随机分为3组,A组种植体周围注射腺病毒Ad5-OPG-EGFP,B组种植体周围注射腺病毒Ad5-EGFP,C组种植体周围注射相同体积的PBS。在植入手术后1周、4周处死动物,行RT-PCR、组织学观察和Micro-CT扫描等检测,采用SPSS15.0软件包对相关数据进行t检验。结果:实验组hOPG表达量显著高于对照组(P<0.05),证实重组腺病毒载体Ad5-OPGEGFP建模成功,可顺利转染到骨组织中并稳定表达。植入术后4周,实验组和对照B组和C组大鼠均有新骨形成、包绕种植体。A组骨面积百分比(%BIC)、骨结合百分比(%BA)、标化后骨体积(BV/TV)、骨结合率(%OI)、单位体积内骨小梁数量(Tb.N)、单位体积内骨小梁厚度(Tb.Th)、骨小梁连接密度(Conn.D)均显著高于B组和C组(P<0.05),单位体积内骨小梁间隔(Tb.Sp)则显著低于B组和C组。结论:hOPG基因转染可在种植体周围稳定表达OPG,加快骨质再生,使种植体骨结合率增加。
OBJECTIVE: To study the role of partially stabilized expression of OPG in the process of bone regeneration and repair around implants. Methods: Sixty-six Sprague-Dawley rats were randomly divided into three groups. Group A was injected adenovirus Ad5-OPG-EGFP around the implant, Group B was injected adenovirus Ad5-EGFP around the implant. Group C was injected with the same volume of PBS . The animals were sacrificed at 1 week and 4 weeks after implantation, and the data were analyzed by RT-PCR, histological observation and Micro-CT scan. SPSS15.0 software package was used for t test. Results: The hOPG expression in the experimental group was significantly higher than that in the control group (P <0.05). The recombinant adenovirus vector Ad5-OPGEGFP was successfully constructed and successfully transfered into bone tissue and stably expressed. Four weeks after implantation, the experimental group and the control group B and C rats were all new bone formation, surrounded by implants. The percentage of bone area (% BIC), percentage of bone union (% BA), BV / TV, bone union rate (% OI), number of trabecular bone per unit volume Tb.Th and Conn.D in unit volume were significantly higher than those in group B and group C (P <0.05), and the ratio of trabecular spacing (Tb.Sp) Was significantly lower than the B group and C group. CONCLUSION: hOPG gene transfection can stably express OPG around the implant, accelerate bone regeneration and increase the rate of implant bone union.
