[Object] To transform endophytic fungus XT5 of T.brevifolia by taxadiene synthase gene.[Method] The taxadiene synthase gene was transferred into T.brevifolia endophytic fungus XT5 by soil agrobacterium-mediated method,and the fermentation yield of taxol was studied.[Result] 8 recombinants were obtained,and the taxol productivity of strain XT5-TS-2 reached as high as 423.983 4 μg/L,53.19% higher than the original strain,which suggested the output of taxol can be improved effectively by taxadiene synthase gene.[Conclusion] The study would provide theoretical foundation for improving the fermentation yield of taxol by T.brevifolia endophytic fungus.
