氯霉素通常可利用其分子结构中的对硝基苯基团,用重氮化法进行含量测定;也可根据其在波长278nm 处有最大吸收(E_(1cm)~(1%)=297.7),用紫外分光光度法定量。但当氯霉素制成制剂时,在生产和贮存过程中,可水解产生对硝基苯基2-氨基1,3-丙二醇(氨基物)和二氯乙酸,氨基物同样具有硝基苯结构,并在波长278nm 处也有吸收,故用重氮化法及紫外分光光度法均不能正确地测定出氯霉素注射
