目的研究NHE3在正常和胆固醇结石患者胆囊黏膜上皮细胞中的表达情况,并探讨NHE3表达异常与结石形成的关系。方法利用Western blot以及免疫组织化学方法检测10例正常与16例胆固醇结石患者胆囊黏膜上皮细胞中NHE3蛋白和亚细胞定位的表达情况。结果 Western blot和免疫组织化学染色方法均显示,与正常胆囊组相比,结石组胆囊黏膜上皮细胞中NHE3表达增高(P<0.05),NHE3同时表达于胆囊黏膜上皮细胞的顶质膜和胞浆内。结论 NHE3表达异常可能与胆囊胆固醇结石发病相关,其发生机理有待进一步研究证实。
Objective To investigate the expression of NHE3 in gallbladder mucosa epithelial cells in patients with normal and cholesterol gallstone and to explore the relationship between NHE3 expression and stone formation. Methods The expression of NHE3 protein and subcellular localization in gallbladder mucosa epithelial cells in 10 normal and 16 patients with cholesterol gallstone were detected by Western blot and immunohistochemistry. Results Western blot and immunohistochemical staining showed that compared with the normal gallbladder group, the expression of NHE3 in the gallbladder mucosa epithelial cells increased (P <0.05), while the expression of NHE3 in the antral and antral cytoplasm of gallbladder epithelial cells Inside. Conclusions The abnormal expression of NHE3 may be related to the pathogenesis of gallstone gallstone in gallbladder. The pathogenesis of NHE3 remains to be confirmed by further study.
