目的:检测妊娠期高血压疾病(妊高征)患者胎盘中神经肽Y(NPY)、内皮素-1(ET-1)及降钙素基因相关肽(CGRP)的水平,观察三者在胎盘中的表达部位并探讨它们在妊高征发病中的作用。方法:用免疫组化法分别检测38例妊高征患者(其中子痫前期轻度20例、重度15例、子痫期3例)及32例正常足月妊娠患者胎盘中NPY、ET-1及CGRP的水平;同时观察NPY、ET-1水平与CGRP的相关关系。结果:妊高征组患者胎盘中NPY、ET-1在滋养细胞和血管内皮细胞都有表达,妊高征组患者胎盘中NPY、ET-1水平高于正常妊娠组患者,差异有统计学意义(P<0.01),而子痫期患者胎盘中NPY、ET-1水平又显著高于子痫前期轻、重度患者,差异有统计学意义(P<0.05);妊高征组患者胎盘中CGRP水平低于正常妊娠组患者,差异有统计学意义(P<0.01)。妊高征组患者胎盘中NPY、ET-1水平与CGRP水平呈负相关性。结论:NPY、ET-1在妊高征组患者胎盘滋养细胞及血管内皮细胞中都有表达;妊高征组患者胎盘中NPY、ET-1水平明显升高,且随着病情的加重,水平逐渐升高。
Objective: To detect the levels of neuropeptide Y (NPY), endothelin-1 (ET-1) and calcitonin gene related peptide (CGRP) in the placenta of patients with gestational hypertension (PIH) In the expression of parts and to explore their role in the pathogenesis of pregnancy-induced hypertension. Methods: Immunohistochemical method was used to detect the expression of NPY and ET-1 in 38 patients with PIH (20 with mild preeclampsia, 15 with severe preeclampsia and 3 with eclampsia) and 32 normal pregnant women And CGRP levels; while NPY, ET-1 levels and CGRP correlation. Results: Placenta of patients with PIH had placental expression of NPY and ET-1 in both trophoblasts and vascular endothelial cells. The levels of NPY and ET-1 in the placenta of PIH patients were higher than those in normal pregnancy group, the difference was statistically significant (P <0.01). The levels of placental NPY and ET-1 in preeclampsia patients were significantly higher than those in mild and severe preeclampsia patients (P <0.05). The levels of CGRP The level was lower than that of normal pregnancy group, the difference was statistically significant (P <0.01). Placental NPY and ET-1 levels in patients with PIH were negatively correlated with CGRP levels. CONCLUSION: NPY and ET-1 are expressed in placental trophoblasts and vascular endothelial cells in PIH patients. The levels of NPY and ET-1 in placenta of patients with PIH are significantly increased, and with the aggravation of disease, Gradually increased.
