目的探讨重组人血管内皮抑素(恩度)联合放疗对人鼻咽癌细胞株1(CNE1)鼻咽癌裸鼠移植瘤的生长及血管内皮生长因子(VEGF)、基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)mRNA表达的影响。方法建立人鼻咽癌细胞裸鼠模型,成瘤后随机分为4组,对照组、恩度组、放疗组和恩度+放疗联合组,分别进行恩度与放疗干预,观察各组移植瘤的生长速度和瘤体大小的变化。4周后处死裸鼠摘取瘤体,应用RT-PCR检测各组瘤体组织中的VECF、MMP-2和MMP-9 mRNA的表达变化。结果联合组对鼻咽癌移植瘤的生长有显著抑制作用,且明显强于其他三组(均P<0.05);联合组中VECF、MMP-2和MMP-9的表达与另外三组相比均显著下调(均P<0.05);VEGF与MMP-2及MMP-9的表达呈正相关(均P<0.01)。结论恩度联合放疗能明显抑制CNE1鼻咽癌移植瘤的生长,恩度对鼻咽癌裸鼠抑制瘤具有放疗增敏作用,这种作用可能与VEGF、MMP-2和MMP-9表达下调有关。
Objective To investigate the effects of recombinant human endostatin combined with radiotherapy on the growth of human nasopharyngeal carcinoma xenografted in human nasopharyngeal carcinoma cell line 1 (CNE1) and the expressions of vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 -2) and matrix metalloproteinase-9 (MMP-9) mRNA expression. Methods The human nasopharyngeal carcinoma cell model was established in nude mice. After tumorigenesis, the mice were randomly divided into 4 groups: control group, Endostar group, radiotherapy group and Endo + radiotherapy combined group. Endostar and radiotherapy intervention were performed respectively. The growth rate and tumor size changes. After 4 weeks, the nude mice were sacrificed and the changes of the expression of VECF, MMP-2 and MMP-9 mRNA in each group were detected by RT-PCR. Results The combination group significantly inhibited the growth of nasopharyngeal carcinoma and was significantly stronger than the other three groups (all P <0.05). Compared with the other three groups, the expression of VECF, MMP-2 and MMP-9 in the combined group (All P <0.05). The expressions of VEGF and MMP-2 and MMP-9 were positively correlated (all P <0.01). Conclusions Endue combined with radiotherapy can significantly inhibit the growth of nasopharyngeal carcinoma xenografts in CNE1. Endeavor radiosensitization of nasopharyngeal carcinoma in nude mice may be related to the down-regulation of VEGF, MMP-2 and MMP-9 .
