目的:观察舒郁散对慢性不可预见性应激(CUMS)抑郁大鼠行为、大脑皮层去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)及突触后致密物95(PSD-95)蛋白表达的影响。方法:50只大鼠随机分为:正常对照组、CUMS模型组、模型加盐酸氟西汀片治疗组、模型加舒郁散小剂量、大剂量治疗组。对大鼠进行21 d的慢性不可预见性应激刺激建立模型,采用糖水实验、悬尾实验观察大鼠行为改变,用ELISA法检测大鼠大脑皮质神经递质的含量,用免疫组织化学染色法观察大鼠大脑皮质PSD-95蛋白表达。结果:盐酸氟西汀组、舒郁散大剂量组大鼠悬尾不动时间明显延长,糖水消耗百分率明显下降,与模型组比差异显著,P<0.01。盐酸氟西汀、舒郁散大剂量组大鼠尾悬不动时间明显减少,糖水消耗量明显增加,与模型组比差异显著,P<0.05。模型组鼠脑组织NE、5-HT的含量显著降低,与正常对照组比差异显著,P<0.05;大剂量舒郁散能显著提高CUMS大鼠脑内NE、5-HT的含量,与模型组比差异显著,P<0.05。模型组大鼠大脑皮层PSD-95蛋白表达减少,与正常组比差异显著,P<0.05;大剂量舒郁散能显著提高CUMS大鼠脑内PSD-95蛋白表达,与模型组比差异显著,P<0.05。结论:动物行为学显示舒郁散具有抗抑郁作用,这种作用与提高慢性应激抑郁大鼠脑内神经递质的含量、提高大脑皮层PSD-95表达有关,上述作用与其剂量呈正相关。
Objective: To observe the effect of Shuyu Powder on behavior, NE, DA, 5-HT and postsynaptic density of rats with chronic unpredictable stress (CUMS) 95 (PSD-95) protein expression. Methods: Fifty rats were randomly divided into normal control group, CUMS model group, model plus fluoxetine hydrochloride tablets treatment group, model plus Shuyu powder low-dose, high-dose treatment group. The rats were subjected to chronic unpredictable stress stimulation for 21 days to establish a model. The behavior of the rats was observed by using the water-swallow test and the tail-suspension test. The content of neurotransmitters in the cerebral cortex of the rats was detected by ELISA. Immunohistochemical staining The cerebral cortex PSD-95 protein expression was observed. Results: The hangover time of fluoxetine hydrochloride group and Shuyu San high dose group were significantly prolonged, and the percentage of sugar consumption significantly decreased compared with the model group (P <0.01). Fluoxetine Hydrochloride and Shuyu San large dose group rats significantly decreased immobility time, consumption of sugar increased significantly, compared with the model group, the difference was significant (P <0.05). Compared with normal control group, the content of NE and 5-HT in model group decreased significantly (P <0.05), while the high dose Shuyu Powder could significantly increase the content of NE and 5-HT in brain of CUMS rats, Group difference was significant, P <0.05. Compared with the model group, the PSD-95 protein expression in the cerebral cortex decreased significantly in the model group (P <0.05), and the difference was significant (P <0.05). The high dose Shuyu Powder could significantly increase the PSD-95 protein expression in the CUMS rat brain, P <0.05. Conclusion: Animal behavior shows that Shuyu San has antidepressant effect, which is related to increasing the level of neurotransmitter in the brain of chronic stress rats and increasing the expression of PSD-95 in cerebral cortex. The above-mentioned effects are positively correlated with the dosage.
