目的在采用相同浓度血管紧张素Ⅱ(AngⅡ)刺激的情况下,研究不同浓度转化生长因子β(TGF-β)抑制剂(Decorin)对体外培养的人脐静脉内皮细胞(HECV)中的Toll样受体2(TLR2)表达的影响。方法取对数生长期的HECV,分为三组:空白对照组、AngⅡ刺激组和AngⅡ+Decorin刺激组,给予药物刺激后采用Western印迹技术检测各组细胞内TLR2蛋白的表达量,同时采用实时定量PCR检测各组细胞内TLR2基因的表达量,并对各组结果进行统计分析。结果 Decorin作用于体外培养的HECV后,该组细胞TLR2的表达量明显高于AngⅡ刺激组但低于正常对照组。结论 TGF-β加强了AngⅡ对HECV-304中TLR2表达的下调作用。
Objective To investigate the effect of different concentrations of Decorin on Toll-like in human umbilical vein endothelial cells (HECV) stimulated with the same concentration of angiotensin Ⅱ (AngⅡ) Receptor 2 (TLR2) expression. Methods HECV in logarithmic growth phase were divided into three groups: control group, Ang Ⅱ stimulation group and Ang Ⅱ + Decorin stimulation group. Western blotting was used to detect the expression of TLR2 protein in each group after stimulation. Quantitative PCR was used to detect the expression of TLR2 gene in each group, and the results of each group were statistically analyzed. Results After Decorin treated HECV in vitro, the expression of TLR2 in the cells was significantly higher than that in the AngⅡ-stimulated group but lower than that in the normal control group. Conclusion TGF-β enhances the down-regulation of TLR2 expression in HECV-304 induced by AngⅡ.
