以秀丽野海棠叶片为外植体诱导愈伤组织并分化出不定芽,得到再生植株,建立了组织培养和快速繁殖体系。结果表明:培养基MS+6-BA 1.0 mg·L~(-1)+2,4-D 0.3 mg·L~(-1)适用于叶片外植体愈伤组织诱导和分化,诱导率为100%,分化率为85.83%;MS+6-BA 1.0 mg·L~(-1)+IBA 0.3 mg·L~(-1)适用于继代增殖培养,平均增殖系数为7.38,且不定芽较粗壮;MS+NAA 0.5mg·L~(-1)+活性炭1 g·L~(-1)适用于生根培养,生根率达到100%;将生长良好的无菌植株进行移栽驯化,存活率达到90%。
The leaves of Begonia officinalis were used as explants to induce callus and differentiate into adventitious buds. The regenerated plants were obtained and tissue culture and rapid propagation system were established. The results showed that MS + 6-BA 1.0 mg · L -1 and 2,4-D 0.3 mg · L -1 were suitable for callus induction and differentiation of leaf explants with the induction rate of 100% and the rate of differentiation was 85.83%. MS + 6-BA 1.0 mg · L -1 and IBA 0.3 mg · L -1 were suitable for subculture, with an average multiplication coefficient of 7.38 and adventitious buds Sturdy.
