目的探讨香丹注射液不同提取部位对麻醉大鼠血流动力的影响。方法将大鼠随机分为7组:生理盐水组、香丹注射液组、香丹注射液石油醚提取部位组(a)、氯仿提取部位组(b)、醋酸乙酯提取部位组(c)、正丁醇提取部位组(d)、水提取部位组(e),给药剂量为16g生药/kg,静脉注射方式给药。用股动脉插管、右颈总动脉心室插管和连续记录肢体Ⅱ导联心电图的方法观察给药前和给药后5、10和15min大鼠的心率(HR)、收缩压(SBP)、舒张压(DBP)、平均压(MAP)、左心室收缩压(LVSP)、左心室终末舒张压(LVEDP)、左心室内压最大上升速率(+dp/dtmax)、左心室开始收缩至dp/dtmax的间隔时间(t-dp/dtmax)和左心室内压最大下降速率(-dp/dtmax)的变化。结果静脉注射相应的受试物后,石油醚提取部位组(a)和乙酸乙酯提取部位组(c)对麻醉大鼠血流动力学的各项指标无显著性影响(P>0.05);氯仿提取部位组(b)能使大鼠SBP、DBP、MAP显著升高(P<0.05);水提取部位组(e)能使大鼠SBP显著升高(P<0.05);正丁醇提取部位组(d)能使大鼠SBP、DBP、MAP、LVSP和-dp/dtmax显著升高(P<0.05);香丹注射液五个不同提取部位对HR、LVEDP、+dp/dtmax和t-dp/dtmax指标无显著性影响(P>0.05)。结论香丹注射液的正丁醇部位提取物具有增强心肌收缩功能和改善心肌舒张功能。
Objective To investigate the effect of different extractive parts of Xiangdan injection on hemodynamics in anesthetized rats. Methods Rats were randomly divided into 7 groups: normal saline group, Xiangdan injection group, Xiangdan injection petroleum ether extraction site group (a), chloroform extraction site group (b), ethyl acetate extraction site group (c) The n-butanol extraction site group (d) and the water extraction site group (e) were administered at a dose of 16 g crude drug/kg and administered intravenously. The heart rate (HR) and systolic blood pressure (SBP) of the rats before and 5, 10, and 15 min after administration were observed using femoral artery catheterization, right common carotid artery ventricular catheterization, and continuous recording of limb II lead electrocardiograms. Diastolic blood pressure (DBP), mean pressure (MAP), left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), maximum rate of increase of left ventricular pressure (+dp/dtmax), left ventricular contraction to dp The interval between /dtmax (t-dp/dtmax) and the maximum rate of decline in left ventricular pressure (-dp/dtmax). Results After intravenous injection of the corresponding test substance, the petroleum ether extract fraction group (a) and ethyl acetate extract fraction group (c) had no significant influence on the hemodynamic indexes of anesthetized rats (P>0.05). Chloroform extraction group (b) can significantly increase the rats SBP, DBP, MAP (P <0.05); water extraction site group (e) can significantly increase the rat SBP (P <0.05); n-butanol extraction Site group (d) significantly increased SBP, DBP, MAP, LVSP, and -dp/dtmax in rats (P<0.05); five different extraction sites of Xiangdan injection to HR, LVEDP, +dp/dtmax, and t -dp/dtmax index had no significant effect (P>0.05). Conclusion The n-butanol extract of Xiangdan injection has enhanced myocardial contractile function and improved myocardial diastolic function.
