目的探讨中药三七提取物人参皂甙-2A体外抗血小板作用。方法取高血压患者及正常人静脉血标本,常规制备富含血小板血浆,分别用不同浓度的硝苯地平、SK&F96365、人参皂甙-2A孵育,以2μmol/L二磷酸腺苷为诱聚药,连续5次测定5min血小板的聚集率,以血小板最大聚集率[maximal platelet aggregation rate,PAG(M)]为观察指标。结果(1)高血压患者PAG(M)为0.89±0.06,与正常人PAG(M) 0.68±0.07比较,差异有显著性(P<0.01);(2)两种浓度(10、20μmol/L)的硝苯地平对正常人和高血压患者PAG(M)均无影响;(3)不同浓度(2.5、5、10、20μmol/L)的SK&F96365对高血压病组的PAG(M)均有抑制作用;(4)不同浓度(2.5、5,10、20μmol/L)的人参皂甙-2A对高血压病组的PAG(M)均有抑制作用(P<0.05,P<0.01);3种浓度(5、10、20μmol/L)的人参皂甙-2A对正常组的PAG(M)均有抑制作用(P<0.05)。结论高血压患者血小板聚集功能明显高于正常人,血小板呈高反应状态,硝苯地平不能抑制血小板聚集;SK&F96365能抑制血小板的聚集功能;人参皂甙-2A能抑制血小板的聚集,有明确的抗血小板作用。
Objective To investigate the antiplatelet effect of ginsenoside-2A extracted from Sanqi, a traditional Chinese medicine. Methods Venous blood samples were collected from hypertensive patients and normal controls. Platelet-rich plasma was routinely prepared and incubated with different concentrations of nifedipine, SK&F96365, and ginsenoside-2A, with 2 μmol/L adenosine diphosphate as the inducer. The platelet aggregation rate of 5 minutes was measured 5 times, and the maximal platelet aggregation rate (PAG(M)) was used as the observation index. Results (1) The PAG (M) of hypertensive patients was 0.89±0.06, compared with normal PAG(M) 0.68±0.07, the difference was significant (P<0.01);(2) Namidipine at two concentrations (10, 20 μmol/L) had no effect on PAG (M) in normal subjects and hypertensive patients; (3) SK&F96365 with different concentrations (2.5, 5, 10, and 20 μmol/L) The PAG(M) of hypertension group was inhibited; (4) Ginsenoside-2A with different concentrations (2.5, 5, 10, and 20 μmol/L) had PAG(M) in hypertension group. Inhibitory effect (P<0.05, P<0.01); Ginsenoside-2A at three concentrations (5, 10, and 20 μmol/L) inhibited the PAG(M) in the normal group (P<0. 05). Conclusions The platelet aggregation function of hypertensive patients is significantly higher than that of normal people. Platelets are highly reactive. Nifedipine does not inhibit platelet aggregation. SK&F96365 inhibits platelet aggregation. Ginsenoside-2A inhibits platelet aggregation and has clear antiplatelet activity. effect.
