目的研究Livinα和Livinβ在儿童急性白血病(childhood acute leukemia,CAL)骨髓单个核细胞(marrowmononuclear cell,MMNC)中的表达与临床意义。方法采用实时PCR(real-time PCR)和Western blot检测Livinα和Livinβ在CAL骨髓单个核细胞中mRNA水平和蛋白质水平的表达。结果Livinα和Livinβ在初诊的39例儿童急性淋巴细胞白血病(acute lymphoblastic leukemia,ALL)和12例急性髓细胞白血病(acute myeloid leukemia,AML)MMNC中的表达率和表达水平均高于对照组(P<0.05),Livinα的表达水平高于Livinβ(P<0.01)。ALL化疗后第19天,幼稚细胞≤5%组Livinα和Livinβ的表达水平较化疗前明显降低(P<0.01)。幼稚细胞>5%组化疗后较化疗前也降低(P<0.05),但化疗后Livinα和Livinβ的表达水平仍明显高于对照组(P<0.01)。结论Livinα和Livinβ在CAL中高表达可能对评估CAL临床危险度及预后有重要意义;提示Livinα和Livinβ可作为观察CAL预后的分子指标之一。
Objective To study the expression and clinical significance of Livinα and Livinβ in bone marrow mononuclear cells (MMNC) of childhood acute leukemia (CAL). Methods The mRNA and protein levels of Livinα and Livinβ in CAL bone marrow mononuclear cells were detected by real-time PCR and Western blot. Results The positive rates of Livinα and Livinβ in 39 cases of acute lymphoblastic leukemia (ALL) and 12 cases of acute myeloid leukemia (MMNC) were higher than those in control group (P <0.05). The expression of Livinα was higher than that of Livinβ (P <0.01). On the 19th day after ALL chemotherapy, the expression of Livinα and Livinβ in naive cells ≤5% was significantly lower than that before chemotherapy (P <0.01). The percentage of naive cells> 5% after chemotherapy was also lower than that before chemotherapy (P <0.05), but the levels of Livinα and Livinβ were still significantly higher than those of the control group after chemotherapy (P <0.01). Conclusion The high expression of Livinα and Livinβ in CAL may be of great significance in assessing the clinical risk and prognosis of CAL. It is suggested that Livinα and Livinβ may serve as one of the molecular markers to evaluate the prognosis of CAL.
