目的:通过观察高、中、低剂量组的清脂胶囊对营养性肥胖大鼠各项指标的影响,进一步明确该药药效及最佳剂量,初步阐明该方抗营养性肥胖的机制。方法:采用高脂乳剂+碳酸饮料+普通饲料混合喂养复制大鼠营养性肥胖模型。各组动物末次给药后,检测血清葡萄糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、瘦素(LEP)、脂联素(ADP)、胰岛素(INS)含量,计算脂肪指数。结果:与模型对照组比较,清脂胶囊高、中剂量组可降低大鼠血清GLU、TG含量,清脂胶囊低剂量组可降低血清TG水平,而清脂胶囊中、低剂量组则显著升高血清HDL-C水平(P<0.05,P<0.01);清脂胶囊高剂量组可降低血清LEP、ADP、INS水平,清脂胶囊中、低剂量组可分别降低ADP、LEP水平(P<0.01)。结论:清脂胶囊高、中、低剂量组均可对抗混合喂养法所致大鼠营养性肥胖的发生,其作用可能与干预肥胖大鼠LEP抵抗,降低血清INS水平有关;综合药效和用药安全度,临床建议采用低剂量作为首选剂量。
OBJECTIVE: To investigate the effects of Qingzhi Capsule on various indexes of nutritional obese rats by observing the effects of high, medium and low doses of Qingzhi Capsules, and further clarify the efficacy and the best dosage of this medicine. Methods: The rat model of nutritional obesity was reproduced by mixed feeding of high fat emulsion + carbonated drink + normal feed. After the last administration, the serum glucose, total cholesterol, triglyceride, low density lipoprotein cholesterol, high density lipoprotein cholesterol, Leptin (LEP), adiponectin (ADP), insulin (INS) content, calculate the fat index. Results: Compared with the model control group, high and medium dose Qingliao capsule can reduce serum GLU, TG content, low dose Qingliao capsule can reduce serum TG levels, and Qingzhi capsule low dose group was significantly increased (P <0.05, P <0.01). The high dose of Qingzhi Capsule could reduce the level of serum LEP, ADP and INS, the level of ADP and LEP in the middle and low dose of Qingzhi capsule could be decreased (P < 0.01). Conclusion: The capsule of high, medium and low dose of Qingzhi Capsule can antagonize the occurrence of nutritional obesity in rats induced by mixed feeding, which may be related to the intervention of LEP resistance in obese rats and the decrease of serum INS level.
