目的建立同时测定杜仲叶中活性成分绿原酸、京尼平苷含量的反相高效液相色谱法。方法采用YMC-packODS-A色谱柱(250mm×4.6mm,5μm),以甲醇-水-冰醋酸(20:80:1.5)为流动相,检测波长237nm,柱温30℃,流速1.0mL.min-1。结果绿原酸在0.0416~0.6240μg内,有较好的线性关系(r=0.9992),绿原酸的加样回收率为102.92%,RSD为1.49%;京尼平苷在0.0816~1.2240μg内,有较好的线性关系(r=0.9989),京尼平苷的加样回收率为102.35%,RSD为1.20%。杜仲叶中绿原酸含量为2.801mg.g-1、京尼平苷含量为0.273mg.g-1。结论所建立的方法简便、准确、可靠,可作为杜仲叶的质量控制方法。
OBJECTIVE To establish an RP-HPLC method for simultaneous determination of chlorogenic acid and geniposid in Eucommia ulmoides leaves. Methods The mobile phase was YMC-pack ODS-A (250 mm × 4.6 mm, 5 μm) with methanol-water-glacial acetic acid (20:80: 1.5) as mobile phase at a detection wavelength of 237 nm with a column temperature of 30 ℃ and a flow rate of 1.0 mL · min -1. Results The linearity of chlorogenic acid was 0.0416 ~ 0.6240μg (r = 0.9992). The recovery rate of chlorogenic acid was 102.92%, RSD was 1.49%. Geniposide was within 0.0816 ~ 1.2240μg , With a good linear relationship (r = 0.9989). The average recovery of geniposide was 102.35% and the RSD was 1.20%. Eucommia leaf chlorogenic acid content of 2.801mg.g-1, geniposide content of 0.273mg.g-1. Conclusion The established method is simple, accurate and reliable and can be used as quality control method of Eucommia ulmoides leaves.
