目的:探讨人颈动脉粥样硬化(AS)时血脂成分,Ⅰ型胶原mRNA及其蛋白与NF-κBmRNA及其蛋白在人颈AS病变中的表达及临床意义。方法:对18例行颈动脉造影确诊有颈AS病变的患者,测定血脂成分,且行颈动脉内膜剥脱术,取出粥样斑块送病检;另取6例意外死亡(排除心血管疾病)的颈动脉标本作为对照;石蜡包埋制成切片。采用免疫组化及原位杂交法染色,检测Ⅰ型胶原mRNA及其蛋白与NF-κBmRNA及其蛋白在AS斑块中表达,并通过CMIAS真彩色医学图像分析系统测出表达的定量A值,进行2组间比较。结果:人颈AS病变中Ⅰ型胶原、NF-κB蛋白及Ⅰ型胶原mRNA、NF-κBmRNA表达定量A值,分别为:0.27±0.08、0.34±0.10及0.49±0.12、0.58±0.15,显著高于对照组(0.10±0.01、0.10±0.02及0.17±0.02、0.18±0.02)的定量表达(P<0·05)。结论:Ⅰ型胶原蛋白、NF-κB蛋白及Ⅰ型胶原mRNA、NF-κBmRNA表达与AS发生、发展有着密切关系,该因素的表达增强是致AS的机制之一。
Objective: To investigate the expression and clinical significance of serum lipid components, type Ⅰ collagen mRNA and NF-κB mRNA and protein in human cervical AS lesions in human carotid atherosclerosis (AS). Methods: Twenty-eight patients with neck AS diagnosed by carotid angiography were enrolled. Blood lipids were measured and carotid endarterectomy was performed. Atherosclerotic plaques were removed for pathological examination. Six other deaths (excluding cardiovascular diseases) Of the carotid artery specimens as a control; paraffin embedded into sections. Immunohistochemistry and in situ hybridization staining, type Ⅰ collagen mRNA and protein and NF-κB mRNA and protein expression in the AS plaque, and through the CMIAS true color medical image analysis system to measure the expression of quantitative A value, Two groups were compared. Results: The quantitative A value of type Ⅰ collagen, NF-κB protein and type Ⅰ collagen mRNA and NF-κB mRNA expression in human cervical AS lesions were 0.27 ± 0.08,0.34 ± 0.10 and 0.49 ± 0.12,0.58 ± 0.15 respectively, which were significantly higher (0.10 ± 0.01, 0.10 ± 0.02 and 0.17 ± 0.02,0.18 ± 0.02) in the control group (P <0.05). CONCLUSION: The expressions of type Ⅰ collagen, NF-κB protein and type Ⅰ collagen mRNA and NF-κB mRNA are closely related to the occurrence and development of AS. The enhancement of this factor is one of the mechanisms responsible for AS.
