为探究胃瘤线虫幼虫体外存活影响因素,观察了黄鳝源胃瘤线虫幼虫在不同温度、溶液、pH值条件下的存活情况。结果表明:胃瘤线虫幼虫在–36℃、–20℃、–7℃的存活时间分别为(2±0.71)h、(5±1.15)h、(5±1.15)d;在4℃、16℃~28℃、32℃及68℃的存活时间分别为(55±5.29)d、9 d以上、3 d及(5±2)s。胃瘤线虫幼虫在pH=7.4、pH=7、pH≥8.87或pH≤5.10溶液中,存活时间分别为(15±2)d、(8±1)d、(2±1)d。胃瘤线虫幼虫在2%葡萄糖溶液、任氏液、0.9%生理盐水、蒸馏水、高浓度(≥6.5%)NaCl溶液中存活时间分别为(14±2)d、(11.67±1.53)d、(10.33±3.21)d、(8±1)d、(4.33±0.58)h。本试验结果可为胃瘤线虫幼虫的实验室保存及肉品安全参数提供依据。
In order to explore the influencing factors of survival of nematode larvae in vitro, we observed the survival of larvae of eel-infected nematode at different temperatures, solutions and pH values. The results showed that the survival time of the nematode larvae at -36 ℃, -20 ℃ and -7 ℃ were (2 ± 0.71) h, (5 ± 1.15) h and (5 ± 1.15) d, respectively. At 4 ℃ and 16 The survival time at ℃ ~ 28 ℃, 32 ℃ and 68 ℃ were (55 ± 5.29) days, 9 days, 3 days and (5 ± 2) days, respectively. The survival time of the nematode larvae was (15 ± 2) days, (8 ± 1) days and (2 ± 1) days in pH = 7.4, pH = 7, pH≥8.87 or pH≤5.10, respectively. The survival time of the nematode larvae was (14 ± 2) d and (11.67 ± 1.53) d in 2% dextrose solution, Ren solution, 0.9% saline, distilled water and high concentration (≥6.5% 10.33 ± 3.21) d, (8 ± 1) d, (4.33 ± 0.58) h. The results of this test may provide the basis for the laboratory preservation and meat safety parameters of the nematode larvae of the stomach.
