肺积方抑制Lewis肺癌肿瘤生长及其抗移植瘤血管生成的实验研究

目的:观察肺积方对移植性肿瘤生长的抑制作用,并探讨其可能的作用机制。方法:将C57小鼠与裸鼠分别随机分为对照组及中药低、中、高剂量组。①建立C57小鼠Lewis肺癌模型,于接种后第10天开始药物干预,中药组灌胃相应浓度的肺积方,对照组予生理盐水。在给药第5、8、11、14天测量小鼠瘤径,灌胃第14天处死小鼠并称取瘤重。②采用Lewis肺癌细胞株接种于裸鼠背部视窗部位,药物干预同上。分别在给药当天及第4、9、14天静脉注射FITC-dextran以显示功能性血管,在激光共聚焦扫描显微镜下测量单位面积的平均血管数量和直径。结果:①在各观察时间节点,高剂量组瘤径均明显小于对照组(P<0.05,P<0.01),而低、中剂量组与对照组无明显差异(P>0.05);第8、11、14天高剂量组瘤径均明显小于低剂量组(P<0.01,P<0.05),第8、11天高剂量组明显小于中剂量组(P<0.01)。14 d后,高剂量组瘤重明显低于对照组(P<0.05);低、中剂量组均低于对照组,但差异无统计学意义(P>0.05)。②第9、14天,中、高剂量组血管数量明显少于对照组与低剂量组(P<0.01),而低剂量组与对照组数量相当(P>0.05);第4天高剂量组血管直径明显小于低剂量组(P<0.05),其余时间点各组血管直径无明显差异(P>0.05)。结论:肺积方对Lewis肺癌小鼠移植瘤的生长具有抑制作用,这种作用可能与减少肿瘤组织中功能性血管的数量,从而抑制肿瘤血管新生有关。 Objective: To observe the inhibitory effect of Fei Ji Fang on the growth of transplanted tumors and to explore its possible mechanism. Methods: C57 mice and nude mice were randomly divided into control group and TCM low, medium and high dose groups. ① C57 mice Lewis lung cancer model was established, and drug intervention was started on the 10th day after inoculation. The rats in the Chinese herb group were given the corresponding prescription of the lung and the control group were given normal saline. On the 5th, 8th, 11th and 14th day after administration, the tumor diameter of the mice was measured. On the 14th day, the mice were sacrificed and the tumor weights were weighed. ② Lewis lung cancer cell lines were inoculated on the back of the window in nude mice, drug intervention ibid. FITC-dextran was intravenously injected on the day of administration and on days 4, 9, and 14 to show functional blood vessels, and the average number of blood vessels per unit area and diameter were measured under a confocal laser scanning microscope. Results: ①The diameter of tumor in the high-dose group was significantly lower than that in the control group at each observation time (P <0.05, P <0.01), while there was no significant difference between the low and medium dose groups and the control group (P> 0.05) The tumor diameter of the high dose group on day 11 and day 14 were significantly lower than that of the low dose group (P <0.01, P <0.05). On the 8th and 11th day, the high dose group was significantly smaller than the medium dose group (P <0.01). After 14 days, the tumor weight of the high-dose group was significantly lower than that of the control group (P <0.05). The low and medium dose groups were lower than the control group, but the difference was not statistically significant (P> 0.05). ② The numbers of blood vessels in the 9th, 14th, middle and high dose groups were significantly less than those in the control group and the low dose group (P <0.01), while those in the low dose group and the control group were similar (P> 0.05) Vascular diameter was significantly lower than the low dose group (P <0.05), the rest of the time point there was no significant difference in vessel diameter (P> 0.05). Conclusion: Feiji Fang can inhibit the growth of transplanted Lewis lung carcinoma in mice, and this effect may be related to reducing the number of functional blood vessels in tumor tissue and thus inhibiting tumor angiogenesis.
