目的:动态观察单侧输尿管结扎(UUO)大鼠肾组织中α-平滑肌肌动蛋白(α-SMA)、E-钙黏蛋白(E-cadherin)的表达及益肾化瘀方对其影响。方法:将清洁级雄性SD大鼠108只随机分为假手术组,模型组,益肾化瘀方高、中、低剂量组及缬沙坦组,除假手术组外各组采用UUO模型,分别于术后第7、14、28天光镜下观察肾间质病理改变,免疫组化法观察大鼠肾组织α-SMA、E-cadherin的表达变化。结果:造模后7d,大鼠肾组织中α-SMA表达即明显增高,而E-cadherin表达逐渐减少;与模型组相比,各治疗组大鼠α-SMA的表达减弱,E-cadherin的表达增强(P<0.01,P<0.05),以益肾化瘀方高剂量组和缬沙坦组变化最显著,二者比较无统计学意义。结论:益肾化瘀方可能通过抑制大鼠肾组织中α-SMA的早期高表达,上调E-cadherin的表达,而发挥抗肾间质纤维化作用。
OBJECTIVE: To dynamically observe the expression of α-SMA and E-cadherin in renal tissues of rats with unilateral ureteral obstruction (UUO) and the effect of Yishen Huayu Fang on it. Methods: 108 male SD rats were randomly divided into sham operation group, model group, Yishen Huayu Fang high, medium and low dose groups and valsartan group. Except sham operation group, UUO model was used in each group, The pathological changes of renal interstitium were observed under the light microscope on the 7th, 14th and 28th day after the operation. The expression of α-SMA and E-cadherin in the kidney were observed by immunohistochemistry. Results: Compared with the model group, the expression of α-SMA in rat renal tissue was significantly increased and the expression of E-cadherin decreased gradually (P <0.01, P <0.05). The changes of Yishen Huayu Recipe high dose group and Valsartan group were the most significant. There was no significant difference between the two groups. Conclusion: Yishen Huayu Recipe may exert anti-renal interstitial fibrosis by inhibiting the early expression of α-SMA in rat renal tissue and up-regulating the expression of E-cadherin.
