用单克隆抗体识别出的分化抗原日益增多,已达CD45。先前的研究已将编码CD6、CD7、CD18和CD45的基因分别定位于人11、17、21和1号染色体。人类p250T细胞活化抗原被B1.19.2单克隆抗体识别,是分子量为250KDa的一条单链多肽抗原,第三次国际人白细胞分化抗原讨论会将其归于CDw26。本文作者用TPA或ConA刺激人外周血淋巴细胞或慢性T淋巴细胞性白血病细胞(T-CLL)与HGPRT缺陷的BE5147细胞(小鼠白血病细胞),用PEG融合,经HAT选择,然后用B 1.19.2抗体借助混合红细胞吸附试验(MHA)筛选抗原表达的阳性孔,最后用有限稀释法克隆阳性孔。结果表明,与B 1.19.2反应的阳性
With monoclonal antibodies recognized by the growing number of differentiation antigens, has reached CD45. Previous studies have mapped the genes encoding CD6, CD7, CD18 and CD45 to human chromosomes 11, 17, 21 and 1, respectively. Human p250T cell activating antigen is recognized by B1.19.2 monoclonal antibody, which is a single-chain polypeptide antigen with a molecular weight of 250KDa. The third international human leukocyte differentiation antigen discussion group attributed it to CDw26. In this study, BE5147 cells (mouse leukemic cells) with human peripheral blood lymphocytes or chronic T-lymphoblastic leukemia cells (T-CLL) and HGPRT deficiencies were stimulated with TPA or ConA, fused with PEG, selected by HAT, and then stimulated with B 1.19 .2 Antibodies Screen positive wells for antigen expression by mixed erythrocyte adsorption assay (MHA), and finally positive wells were cloned by limiting dilution.
