目的探讨硝酸钇对生长发育期大鼠肾脏的影响。方法实验分0、20、80和320 ppm(以饲料中钇计)共四组,仔鼠从GD0持续暴露至PND70,每组取20只仔鼠(雌雄各半)的肾脏检测抗氧化酶活性、miRs表达水平、肾组织病理学和KIM-1抗原表达水平的变化。结果与对照组比较,雌鼠320 ppm组miR-18a-5p、miR-21-3p降低;雄鼠20 ppm组miR-18a-3p降低,320 ppm组升高,320 ppm组miR-18a-5p、miR-155-5p升高;雌鼠80 ppm组GSH-Px活性升高,320ppm组GSH-Px活性降低,320 ppm组GST活性降低;雌、雄鼠320 ppm组KIM-1表达水平升高。结论 GD0至PND70硝酸钇暴露,320 ppm组大鼠肾抗氧化能力降低,影响miRs和KIM-1表达水平,提示生命早期硝酸钇暴露可能通过降低抗氧化能力影响肾功能。
Objective To investigate the effect of yttrium nitrate on the growth and development of rat kidney. Methods The experiment was divided into four groups at 0, 20, 80 and 320 ppm (calculated as yttrium in feed). The pups were continuously exposed from GD0 to PND70. Twenty kidneys (male and female) in each group were tested for their antioxidant enzyme activities , MiRs expression, renal histopathology and KIM-1 antigen expression levels. Results Compared with the control group, miR-18a-5p and miR-21-3p in the 320 ppm group of the female rats were decreased. The levels of miR-18a-3p in the 20 ppm group and miR-18a-3p , MiR-155-5p increased; the activity of GSH-Px in 80 ppm group increased, the activity of GSH-Px in 320 ppm group decreased, while the activity of GST decreased in 320 ppm group; the expression level of KIM-1 in 320 ppm group increased . Conclusion GD0 to PND70 yttrium nitrate exposure, 320 ppm group decreased renal antioxidant capacity, affecting miRs and KIM-1 expression levels, suggesting that early life yttrium nitrate exposure may affect the renal function by reducing the antioxidant capacity.
