目的:分析超极化激活环核苷酸调控通道基因家族-2(HCN2)基因及超极化激活环核苷酸调控通道基因家族-4(HCN4)基因在风湿性心脏病二尖瓣狭窄(风心二狭)患者并发心房颤动中的协同表达作用,探讨其并发心房颤动的机制。方法:收集43例风心二狭患者右心耳组织,实验组为并发心房颤动患者(AF组),对照组为窦性心律患者(SR组)。采用逆转录聚合酶链反应(RT-PCR)及荧光定量PCR(FQ-PCR)检测HCN2及HCN4的表达,分析两者的相关性。结果:RT-PCR及FQ-PCR结果均显示,AF组HCN2及HCN4表达量均高于SR组(P<0.01),AF组HCN2与HCN4表达上调率均呈现正相关(r1=0.697,P1<0.01;r2=0.659,P2<0.01)。结论:HCN2与HCN4的协同表达作用在风湿性心脏病二尖瓣狭窄患者并发心房颤动的发生发展中起重要的作用。
OBJECTIVE: To analyze the relationship between HCN2 gene and hyperonuclear activation-regulated HCN4 gene in patients with rheumatic mitral stenosis Wind and heart two narrow) patients with atrial fibrillation in the co-expression of the role of its mechanism of concurrent atrial fibrillation. Methods: The right atrial appendage was collected from 43 patients with wind-heart and two-stenosis. The experimental group was atrial fibrillation (AF group) and the control group was sinus rhythm (SR group). The expression of HCN2 and HCN4 were detected by reverse transcription-polymerase chain reaction (RT-PCR) and real-time quantitative PCR (FQ-PCR) Results: The expression of HCN2 and HCN4 in AF group was significantly higher than that in SR group (P <0.01), and the up-regulation rates of HCN2 and HCN4 in AF group were positively correlated (r1 = 0.697, P1 < 0.01; r2 = 0.659, P2 <0.01). Conclusion: The synergistic expression of HCN2 and HCN4 plays an important role in the occurrence and development of atrial fibrillation in patients with rheumatic mitral stenosis.
