目的探讨搜剔肺络方对肺间质纤维化大鼠的作用及可能机制。方法 128只SD大鼠随机分为假手术组、模型组、中药组、西药组各32只;除假手术组外,其余各组一次性气管内滴注盐酸博莱霉素制备肺纤维化大鼠模型,假手术组气管内注射等体积生理盐水,造模后24h中药组给予搜剔肺络方0.68g/(kg·d)灌胃,西药组给予泼尼松龙5mg/(kg·d)灌胃,模型组和假手术组给予等体积的去离子水,每日1次,分别于第7、14、28、45天处死,取肺组织行病理学观察肺泡炎程度和肺纤维化程度,检测血清转化生长因子-β1(TGF-β1)含量及肺组织Smad3、Smad7蛋白的表达。结果与模型组比较,第7、14、45天中药组和西药组肺泡炎程度均明显减轻(P<0.05),第7、45天中药组和西药组肺纤维化程度明显减轻(P<0.05)。与模型组比较,中药组和西药组TGF-β1含量差异无统计学意义(P>0.05);各时间点中药组和西药组Smad3均较模型组明显减少(P<0.05),且中药组明显低于西药组(P<0.05);各时间点中药组和西药组Smad7较模型组均明显增加(P<0.05)。结论搜剔肺络方对博莱霉素致大鼠肺纤维化有一定的抑制作用,其机制可能与调节Smad信号表达有关。
Objective To investigate the effect and possible mechanism of Xunxieluo Liaofang on pulmonary interstitial fibrosis in rats. Methods Thirty-two SD rats were randomly divided into sham operation group, model group, TCM group and western medicine group, with 32 rats in each group. In addition to the sham-operation group, all the other groups were given intratracheal instillation of bleomycin hydrochloride Rats were injected intraperitoneally with equal volume of saline for sham-operation group. The Chinese medicine group was given 0.24 g / (kg · d) gavage at 24h after modeling, while the western medicine group was given prednisolone 5 mg / (kg · d ) Gavage, model group and sham operation group were given an equal volume of deionized water once a day, were killed on the 7th, 14th, 28th and 45th day respectively. The pathological examination of lung tissue was used to observe the degree of alveolitis and pulmonary fibrosis The level of serum TGF-β1 and the expression of Smad3, Smad7 protein in lung tissue were detected. Results Compared with the model group, the degree of alveolitis in the Chinese medicine group and the western medicine group were significantly reduced on the 7th, 14th and 45th days (P <0.05), and on the 7th and 45th days, the degree of pulmonary fibrosis in the traditional Chinese medicine group and the western medicine group was significantly reduced (P <0.05 ). Compared with model group, the content of TGF-β1 in Chinese medicine group and western medicine group had no significant difference (P> 0.05); Smad3 in Chinese medicine group and Western medicine group decreased significantly (P <0.05) at each time point, (P <0.05). At each time point, the Smad7 levels of Chinese medicine group and western medicine group were significantly higher than those of the model group (P <0.05). CONCLUSION: Choi Fei Liao Fang can inhibit pulmonary fibrosis induced by bleomycin in rats, which may be related to the regulation of Smad signaling.
