为了研究可供人体应用的造血干细胞动员剂,将低相对分子质量葡聚糖硫酸酯(DS)给狗静脉注射,观察其对造血干细胞的动员作用。结果显示,注射DS15mg/kg1h后外周血中WBC、单个核细胞(MNC)和粒-巨细胞集落形成单位(CFU-GM)计数开始升高,3h达到峰值,每升血中的细胞计数分别为药前的1.6、2.1和4.5倍。外周血中WBC、MNC计数,随着DS剂量增加而增加,注射DS60mg/kg时WBC、MNC数分别为药前的2.1和4.0倍。注射不同剂量DS〈以15mg/kg或30mg/kg组动物外周血中各系祖细胞升高幅度最大,CFU-GM、CFU-Meg、红系爆增式集落形成单位(BFU-E)和CFU-Mix每升血中计数分别为药前的9.0、7.2、10.3和13.0倍
In order to study human hematopoietic stem cell mobilization agent, low molecular weight dextran sulfate (DS) was intravenously injected into dogs to observe its mobilization on hematopoietic stem cells. The results showed that the counts of WBC, mononuclear cells (MNC) and granulocyte-macrophage colony forming units (CFU-GM) in peripheral blood began to increase after the injection of DS15mg / kg for 1h, reached the peak at 3h, 1.6, 2.1 and 4.5 times before the drug. The counts of WBC and MNC in peripheral blood increased with the increase of DS dose. The numbers of WBC and MNC at injection of DS60mg / kg were 2.1 and 4.0 times higher than those before treatment, respectively. In the group of different dosages of DS <15mg / kg or 30mg / kg, the peripheral blood progenitor cells showed the highest increase in the range of CFU-GM, CFU-Meg, erythrocytic burst forming colony forming units (BFU- -Mix per liter of blood count were 9.0, 7.2,10.3 and pre-drug 13.0 times
