目的:探讨大肠癌CD44v6表达、p(53)基因突变和染色体倍性与大肠癌转移之间的关系和作用机理。方法:流式细胞仪测定大肠癌细胞倍性及其在细胞周期各相中的分布;RT-PCR方法及特异探针D3对大肠癌细胞进行Southern Blot,并对杂交条带进行辉度扫描;银染PCR单链构象多态性(SSCP)技术检测大肠癌细胞p(53)基因突变。结果:正常对照、良性腺瘤、肿瘤未转移和肿瘤转移组其CD44v6阳性率和p(53)基因突变率呈逐渐上升趋势,CD44v6辉度扫描表明肿瘤未转移组(2317.26±198.73)和转移组(10024.16±855.40)相比较,表达量有显著差异(P<0.02);CD44v6阳性和阴性两组中G2M期细胞均值分别占6.24%和5.48%(P>0.05),异倍体细胞分别为62.16%和60.00%(P>0.5);而p(53)基因突变和未突变两组中G2M细胞占10.33%和4.01%(P<0.05),异倍体细胞分别为85.20%和30,00%(P<0.005);肿瘤异倍体细胞转移率显著高于二倍体细胞。结论:CD44v6的表达与转移高度相关,CD44v6和p(53)基因突变与肿瘤转移关系可能涉及不同的作用机理。C?
Objective: To investigate the relationship between CD44v6 expression, p(53) gene mutation and ploidy of colorectal carcinoma and metastasis of colorectal cancer and its mechanism of action. Methods: Flow cytometry was used to measure the ploidy of colorectal cancer cells and their distribution in each phase of the cell cycle. RT-PCR method and specific probe D3 were used for Southern Blot of colorectal cancer cells, and the hybridization bands were scanned by luminance. Silver staining of single-stranded conformation polymorphisms (SSCP) technique for the detection of p (53) mutations in colorectal cancer cells. RESULTS: The positive rate of CD44v6 and the mutation rate of p(53) gene in the normal control, benign adenoma, tumor non-metastasis and tumor metastasis group showed a gradually increasing trend. The CD44v6 luminance scan showed no tumor metastasis group (2317.26±198.73). Compared with the metastatic group (10024.16±855.40), the expression levels were significantly different (P<0.02); the mean values of G2M phase cells in the CD44v6 positive and negative groups were 6.24% and 5.48, respectively. % (P>0.05), aneuploid cells were 62.16% and 60.00% respectively (P>0.5); while p(53) gene mutations and non-mutation were the two groups. 33% and 4.01% (P<0.05). Aneuploid cells were 85.20% and 30.00%, respectively (P<0.005). The metastasis rate of tumor aneuploid cells was significantly higher than twice. Somatic cells. Conclusion: The expression of CD44v6 is highly correlated with metastasis. The relationship between CD44v6 and p(53) mutations and tumor metastasis may involve different mechanisms. C?
