目的研究不同活化状态的巨噬细胞与雪旺细胞株(RSC96)共培养条件下雪旺细胞神经营养因子(NGF)及层粘连蛋白(Laminin,LN)的表达情况。方法利用Transwell建立大鼠腹膜腔巨噬细胞和RSC96细胞的共培养体系,分别将RSC96细胞与未激活巨噬细胞、激活态巨噬细胞共培养;采用Real-time PCR检测、Western blot分析、以及酶联ELISA测定等研究手段,比较各组NGF及LN的表达差异。结果与未激活巨噬细胞共培养组RSC96细胞NGF表达增强,激活态组表达明显增强;而各组LN表达差异不明显。结论巨噬细胞对RSC96细胞NGF表达有促进作用,受其活化状态的影响,激活状态的巨噬细胞的促进作用更明显;对影响RSC96细胞迁移功能的LN的表达作用不明显,无统计学意义,有待进一步研究。
Objective To investigate the expression of nerve growth factor (NGF) and laminin (LN) in Schwann cells co-cultured with different activated macrophages and Schwann cell line (RSC96). Methods Transwell was used to establish a co-culture system of peritoneal macrophages and RSC96 cells. RSC96 cells were co-cultured with non-activated macrophages and activated macrophages, respectively. Real-time PCR and Western blot analysis Enzyme-linked immunosorbent assay (ELISA) and other research methods to compare the expression of NGF and LN in each group. Results Compared with non-activated macrophages, the expression of NGF in RSC96 cells was enhanced and the expression in activated group was significantly increased. However, the LN expression in each group was not significantly different. CONCLUSION Macrophages promote the expression of NGF in RSC96 cells. The activation of macrophages by activated macrophages is more obvious under the influence of its activation state. The expression of LN in RSC96 cells is not obvious and has no statistical significance , Pending further study.
