目的:观察高糖环境下豚鼠膀胱Cajal间质细胞(interstitial cells of Cajal,ICCs)超微结构,探讨糖尿病膀胱(diabetic cystopathy,DCP)的发病机制。方法:采用酶消化法原代培养,透射电镜观察5、10、15mol/L葡萄糖浓度下培养24、72h的ICCs超微结构。结果:随糖浓度增高培养时间延长,ICCs内细胞器明显减少,线粒体大小不等,数量减少,肿胀、空泡样改变甚至溶解,内质网扩张、空泡样变,胞质广泛溶解,突起消失。结论:高糖环境对豚鼠膀胱ICCs超微结构造成显著影响,这种结构破坏可能是造成DCP发病的重要原因之一。
OBJECTIVE: To observe the ultrastructure of interstitial cells of Cajal (ICCs) in guinea pig bladder under high glucose condition and to explore the pathogenesis of diabetic cystopathy (DCP). Methods: The primary culture of the ICCs was induced by enzyme digestion. The ultrastructures of ICCs cultured for 24 and 72 hours were observed under transmission electron microscope at 5, 10, 15mol / L glucose concentration. Results: With the increase of glucose concentration, the number of organelles decreased, the number of organelles decreased, the number of mitochondria decreased, the number of organelles changed or even dissolved, the endoplasmic reticulum was dilated, the vacuoles changed, and the cytoplasm was widely dissolved. . Conclusion: The hyperglycemic environment has a significant effect on the ultrastructure of bladder ICCs in guinea pigs. This structural disruption may be one of the important causes of DCP.
