目的 :分析中国临床分离的幽门螺杆菌 (Hp)的cag致病岛的差异和不同激酶抑制剂对Hp诱导中国人胃上皮细胞IL - 8分泌的影响。方法 :在体外分别用中国临床分离的cagA+cagE+、cagA+cagE-、cagA-cagE-的Hp与中国人胃上皮细胞MGC - 80 3共同培养 ,IL - 8分泌用ELISA进行检测 ,比较蛋白激酶A、C、G和蛋白酪氨酸激酶的抑制剂对Hp诱导胃上皮细胞IL - 8分泌的影响。结果 :cagA+cagE+Hp显著增加了胃上皮细胞IL - 8的分泌 ,cagA+cagE-、cagA-cagE+Hp作用次之 ,而ca gA-cagE-Hp不能增加胃上皮细胞IL - 8的分泌。蛋白激酶A、C、G的抑制剂阻断Hp增加胃上皮细胞IL - 8的分泌 ,而蛋白酪氨酸激酶的抑制剂不能阻断了Hp增加胃上皮细胞IL - 8的分泌。结论 :cagA+cagE+Hp显著增加了胃上皮细胞IL - 8的分泌并且依赖于蛋白酪氨酸激酶的磷酸化
OBJECTIVE: To analyze the difference of cag pathogenicity isolates from clinically isolated Helicobacter pylori (Hp) in China and the effects of different kinase inhibitors on Hp - induced IL - 8 secretion in Chinese human gastric epithelial cells. Methods: Human gastric epithelial cells MGC - 803 were co - cultured with cagA + cagE +, cagA + cagE- and cagA-cagE- respectively isolated from Chinese in vitro. The secretion of IL - 8 was detected by ELISA. The protein kinase Effect of Inhibitors of A, C, G and Protein Tyrosine Kinase on Hp - induced IL - 8 Secretion in Gastric Epithelial Cells. Results: cagA + cagE + Hp significantly increased secretion of IL - 8 in gastric epithelial cells, followed by cagA + cagE- and cagA-cagE + Hp, while ca gA-cagE-Hp did not increase secretion of IL - 8 in gastric epithelial cells . Inhibitors of protein kinases A, C and G blocked Hp and increased secretion of IL - 8 in gastric epithelial cells. However, inhibitors of protein tyrosine kinase blocked Hp and increased secretion of IL - 8 in gastric epithelial cells. CONCLUSION: cagA + cagE + Hp significantly increases the secretion of IL - 8 in gastric epithelial cells and relies on the phosphorylation of protein tyrosine kinase
