目的探讨金雀异黄素对人红白血病细胞系K562细胞的生长抑制、诱导凋亡作用。方法不同浓度的金雀异黄素处理K562细胞后,用MTT比色法检测细胞生长增殖作用,Wright-Gimesa法观察细胞的形态学变化,FCM检测K562细胞凋亡峰。结果金雀异黄素对K562细胞具有与时间、浓度呈正相关的生长抑制作用。形态学显示金雀异黄素高浓度组细胞发生核固缩、核碎裂,并有凋亡小体出现。细胞周期显示对照组K562 G2/M期细胞8.9%,金雀异黄素75·mol·L·1时升高到60.6%(P<0.05);金雀异黄素75·mol·L-1时K562细胞亚G1期细胞为27.3%,对照组为0.4%(P<0.05)。结论金雀异黄素对K562细胞具有很强的生长抑制作用,K562细胞可发生明显G2/M期阻滞,金雀异黄素诱导了K562细胞发生凋亡。
Objective To investigate the effects of genistein on the growth of human erythroleukemia cell line K562 and the induction of apoptosis. Methods K562 cells were treated with genistein at different concentrations. MTT assay was used to detect the proliferation of K562 cells. Wright-Gimesa method was used to observe the morphological changes of K562 cells. The apoptotic peak of K562 cells was detected by FCM. Results Genistein had a growth-inhibitory effect on K562 cells in a time-and concentration-dependent manner. Morphology showed that genistein at high concentration had nuclear pyknosis and nuclear fragmentation, and apoptotic bodies appeared. The cell cycle showed that the cell cycle of K562 G2 / M phase was 8.9% in control group and 60.6% (P <0.05) when genistein was 75 mol·L · 1, while the genistein 75 mol·L -1 The percentage of cells in sub-G1 phase in K562 cells was 27.3% and that in control group was 0.4% (P <0.05). Conclusions Genistein has a strong growth inhibitory effect on K562 cells. G2 / M arrest in K562 cells can be observed. Genistein induces apoptosis in K562 cells.
