目的探讨凋亡抑制蛋白生存素、抑癌基因p16在骨肉瘤中的表达与骨肉瘤的生物学特性的关系。方法采用免疫组化SP法,检测52例骨肉瘤、15例骨软骨瘤及15例正常骨质标本的生存素蛋白和p16蛋白表达。结果生存素蛋白在骨肉瘤中的表达显著高于骨软骨瘤和正常骨质(P<0.05),在肿瘤的侵袭转移方面差异有显著性(P<0.05),而在性别、年龄、肿瘤直径、病理类型、病理分级及临床分期等方面差异无显著性(P>0.05)。p16蛋白在骨肉瘤中的表达显著低于骨软骨瘤和正常骨质(P<0.05),在肿瘤的病理分级方面差异有显著性(P<0.05),而在性别、年龄、肿瘤直径、病理类型、侵袭转移及临床分期等方面差异无显著性(P>0.05)。在骨肉瘤p16蛋白表达阳性、阴性组中生存素蛋白表达的阳性率分别为73.9%和66.7%(χ2=0.142,P=0.707),经统计学分析,生存素蛋白和p16蛋白表达之间无明显相关(r=0.094,P=0.509)。结论凋亡抑制蛋白生存素在侵袭转移的骨肉瘤组呈高表达;抑癌基因p16在骨肉瘤中随着病理分级的降低,阳性表达率有降低趋势。提示凋亡抑制蛋白生存素、抑癌基因p16在骨肉瘤的发生和发展的不同环节中起重要作用,测定生存素蛋白表达可作为预测预后的重要参考指标,测定p16蛋白表达可作为判断骨肉瘤恶性程度的重要参考指标,并将为肿瘤的基因治疗提供新的靶点。
Objective To investigate the relationship between the expression of survivin, inhibitor of oncogene p16 and the biological characteristics of osteosarcoma in osteosarcoma. Methods Immunohistochemical SP method was used to detect the expression of survivin and p16 protein in 52 cases of osteosarcoma, 15 cases of osteochondroma and 15 cases of normal bone. Conclusions Apoptosis protein survivin is highly expressed in the invasion and metastasis osteosarcoma group. The expression of tumor suppressor gene p16 in osteosarcoma decreases with the decreasing of pathological grade. It is suggested that the inhibitor of apoptosis protein p16 and tumor suppressor gene p16 play an important role in the different stages of the development and progression of osteosarcoma. Survivin protein expression may play an important role in predicting the prognosis of patients with osteosarcoma. To determine the expression of p16 protein, An important reference index of malignancy, and will provide a new target for the gene therapy of tumors.
