目的:建立测定蛹虫草子实体正丁醇部位腺苷含量的高效液相色谱(HPLC)分析方法,并在此基础上初步建立正丁醇部位的特征图谱。方法:利用Angilent TC-C18(250mm×4.6mm,5μm)色谱柱,甲醇5%~30%为流动相梯度洗脱,分析时间60min,检测波长260nm,流速1.0mL·min-1,柱温25℃;对样品提取条件,萃取条件,流动相,检测波长等进行了系统优化设计。结果:测定了11个批次的蛹虫草正丁醇部位样品腺苷的含量平均值为0.536%,初步构建了蛹虫草子实体正丁醇部位HPLC特征图谱。结论:该方法准确简便,重复性好,可作为控制蛹虫草子实体正丁醇提取物内在的质量标准。
Objective: To establish a high performance liquid chromatography (HPLC) method for the determination of adenosine in the n-butanol fraction of Cordyceps militaris fruiting body. Based on this, a preliminary characterization of the n-butanol fraction was established. METHODS: The mobile phase was eluted with 5% ~ 30% methanol on a Angilent TC-C18 column (250 mm × 4.6 mm, 5 μm). The analysis time was 60 min, the detection wavelength was 260 nm, the flow rate was 1.0 mL · min- ℃; the sample extraction conditions, extraction conditions, mobile phase, detection wavelength, etc. were optimized system design. Results: The average content of adenosine in n-butanol fraction of 11 batches of Cordyceps militaris was determined to be 0.536%. The HPLC profile of n-butanol fraction of Cordyceps militaris was preliminarily established. Conclusion: The method is accurate, simple and reproducible, and can be used as an intrinsic quality standard for controlling the n-butanol extract of Cordyceps militaris.
