以苹果果肉为试材,通过比较总蛋白不同提取方法 (TCA-丙酮沉淀法、匀浆法以及改良酚抽提法),建立适于苹果果肉总蛋白提取技术。从所得双向电泳图谱分离效果来看,改良酚抽提法所得蛋白点个数最多、图谱背景清晰,分离效果好,明显优于TCA-丙酮沉淀法和匀浆法。同时,通过对各图谱分离效果的综合比较,认为在现有研究条件下,苹果果肉总蛋白双向电泳分析的理想上样量为250μg。
Using apple pulp as test material, the total protein extraction technology for apple pulp was established by comparing different total protein extraction methods (TCA-acetone precipitation method, homogenization method and modified phenol extraction method). From the results of the two-dimensional electrophoresis separation results, modified phenol extraction obtained the largest number of protein spots, clear background of the map, the separation effect is good, significantly better than the TCA-acetone precipitation and homogenization. At the same time, through the comprehensive comparison of each spectrum separation effect, it is considered that the ideal loading capacity of two-dimensional electrophoresis analysis of apple pulp total protein is 250μg under the existing research conditions.
