用白细胞介素6(interleukin6,IL6)依赖细胞株B9的MTT法,研究了4种刺激剂PMA,A23187,fMLP和LPS对小鼠腹腔巨噬细胞生成及分泌IL6的影响。分别检测了巨噬细胞培养上清液及胞溶部分IL6的含量。结果表明,PMA(01~10μmol·L-1),A23187(001~1μmol·L-1),fMLP(0025~25μmol·L-1)均能促进巨噬细胞生成及分泌IL6,且有量效关系。LPS(01~10μg·ml-1)能增加巨噬细胞培养上清液中IL6的含量,但对胞溶部分无影响。结果提示:巨噬细胞IL6的生成及分泌与蛋白激酶C(PKC)和钙离子通道活化有关;LPS与其他3种刺激剂对胞溶部分IL-6生成的不同作用提示可能存在不同作用机制
The effects of four stimulators PMA, A23187, fMLP and LPS on the production and secretion of IL6 by peritoneal macrophages were studied by MTT assay of interleukin 6 (IL6) dependent cell line B9. Were detected macrophage cell culture supernatant and cytolytic IL 6 content. The results showed that PMA (01 ~ 10μmol·L-1), A23187 (001 ~ 1μmol·L-1) and fMLP (0025 ~ 25μmol·L-1) all promoted macrophage formation And secretion of IL 6, and the dose-effect relationship. LPS (0 1 ~ 10μg · ml-1) can increase the content of IL6 in macrophage culture supernatant, but had no effect on the cell lysate. The results suggest that the production and secretion of IL-6 by macrophages are related to the activation of protein kinase C (PKC) and calcium channels. The different effects of LPS and other three stimulators on the production of IL-6 may indicate different effects mechanism
