目的:应用基因表达谱芯片对糖尿病与正常大鼠种植体周围骨组织的基因表达谱进行对比分析,并筛选差异基因。方法:将6只Wistar大鼠随机分为糖尿病组和正常组,糖尿病组采用链脲佐菌素(Streptozotocin,STZ)腹腔注射法建立糖尿病大鼠模型。分别在大鼠右侧胫骨骨骺端植入钛种植体,3个月后,截取种植体周围骨组织,提取总RNA,进行包括17983个基因的全基因组表达谱芯片检测,对差异表达基因进行GO分析,并采用实时定量PCR对芯片结果进行验证。结果:芯片结果显示,差异基因共1084个,其中糖尿病组表达增强352个,表达降低732个,GO分析涉及1154个不同的功能分类。结论:糖尿病大鼠种植体周围骨组织中与成骨相关的基因表达降低,与脂类代谢相关的基因表达升高。
OBJECTIVE: To compare and analyze gene expression profiles of bone tissue around diabetic and normal rats using gene expression microarray and to screen differential genes. Methods: Six Wistar rats were randomly divided into diabetic group and normal group. Diabetic rats were induced by streptozotocin (STZ) intraperitoneal injection to establish diabetic rat model. Titanium implants were implanted into the right tibial metaphysis of rats respectively. Three months later, the bone tissue around the implant was intercepted and total RNA was extracted for genome-wide expression profiling of 17,983 genes. The differentially expressed genes were subjected to GO Analysis, and using real-time quantitative PCR to verify the chip results. Results: The results of the chip showed that there were 1084 differential genes, including 352 in the diabetic group, 732 in the diabetic group, and 1154 in the GO analysis. CONCLUSION: Osteogenesis-related gene expression is decreased in the bone around the implant and the gene expression associated with lipid metabolism is increased in diabetic rats.
