目的:评价增免升白冲剂的药效,探讨升高白细胞作用机理。方法:腹腔注射CP和~(60)Co照射LACA小鼠复制WBC减少模型,并经口给予不同剂量的该药治疗9d后计数WBC、BMC、CFU-S、CFU-GM并测定BMC中DNA含量。结果:1500mg·kg~(-1)治疗9d CP和~(60)Co模型小鼠WBC计数均恢复至正常,CP模型小鼠CFU-S和DNA含量亦恢复正常;3000mg·kg~(-1)治疗9d,两种模型小鼠各项检测指标均恢复至正常水平,疗效明显优于鲨肝醇;21例白细胞减少症志愿受试者按100mg·kg-1日剂量连续治疗30d,其中20例WBC计数均升至4×10~9·L~(-1)以上。结论:增免升白冲剂有明显的升高WBC、BMC、CFU-S、CFU-GM计数和增加DNA含量的作用,其疗效明显优于鲨肝醇,其机理可能是组方中各有效成分有改善造血微环境,促进DNA合成,促进造血干细胞增殖、分化和成熟作用。
OBJECTIVE: To evaluate the efficacy of Zengji Shengbai Granules and to explore the mechanism of increasing leukocyte activity. METHODS: Intraperitoneal injection of CP and ~(60)Co irradiated LACA mice were used to replicate the WBC reduction model, and after oral administration of different doses of the drug, WBC, BMC, CFU-S, CFU-GM were counted and the DNA content in BMC was measured after 9 days. . RESULTS: The WBC counts of mice treated with 1500 mg·kg -1 for 9 days CP and 60 Co were all restored to normal, and the CFU-S and DNA contents in CP mice also returned to normal. 3000 mg·kg -1 9 days after treatment, the detection indexes of the two mouse models recovered to normal levels, and the efficacy was significantly better than shark hepatol; 21 patients with leukopenia volunteers were treated with a daily dose of 100 mg·kg-1 for 30 days, of which 20 The number of cases of WBC rose to 4 × 10 ~ 9 · L -1 or more. Conclusion: Zengji Shengbai Granules can obviously increase the counts of WBC, BMC, CFU-S, CFU-GM and increase DNA content. Its curative effect is obviously superior to shark liver alcohol, and the mechanism may be the active ingredients in the formula. It can improve the hematopoietic microenvironment, promote DNA synthesis, and promote the proliferation, differentiation and maturation of hematopoietic stem cells.
