目的 :为四君子汤免疫药理作用的物质基础研究提供资料。方法 :以多糖对小鼠溶血素抗体生成反应的影响为药效评价指标 ,比较四君子汤复方多糖免疫活性部位 (SJZPS V)各组分 (SJZPS Va~d)的免疫活性 ;应用凝胶过滤法 ,自免疫活性最强的组分SJZPS Vb中分离得到 2个酸性多糖SJZPS Vb 1~ 2 ,分别采用比色法、凝胶过滤法、甲基化分析、GC、GC/MS联用等技术 ,进行了理化性质研究。结果 :SJZPS Vb 1~ 2的分子量分别为 3 83及2 6 0万 ;组成单糖及其分子摩尔比分别为 :SJZPS Vb 1glucose :galactose :mannose 1∶0 4 6∶2 15 ;SJZPS Vb 2 glucose:galactose :mannose 1∶1 4 1∶4 18。甲基化分析结果表明 ,SJZPS Vb 1及SJZPS Vb 2的组成单糖中 ,末端、4 、6 、4或 6位分支的甘露糖残基数目基本相当 ,而半乳糖的连接位点则有较大差别。结论 :SJZPS Vb 1及SJZPS Vb 2的免疫活性有待研究 ,其结构的异同与活性有否相关值得研究。
Objective : To provide material for the basic research on the immune pharmacology of Sijunzi Decoction. Methods: The effect of polysaccharides on the antibody production of hemolysin in mice was evaluated as an index of drug efficacy. The immune activity of SJZPS V (SJZPS Va) was compared with that of Sijunzi Decoction Polysaccharide polysaccharides (SJZPS V). Gel filtration was used. Two acidic polysaccharides, SJZPS Vb 1~2, were isolated from SJZPS Vb, the most potent component of immune activity, using colorimetric, gel filtration, methylation analysis, GC, and GC/MS combination techniques, respectively. Physical and chemical properties were studied. RESULTS: The molecular weights of SJZPS Vb 1~2 were 3 83 and 26000, respectively. The composition of monosaccharides and their molar ratios were: SJZPS Vb 1glucose :galactose :mannose 1∶0 4 6∶2 15 ; SJZPS Vb 2 glucose :galactose :mannose 1:1 4 1:4 18. The results of methylation analysis showed that the number of mannose residues at the terminal, 4, 6, 4, or 6 branches of SJZPS Vb 1 and SJZPS Vb 2 were basically the same, while the galactose binding sites were comparable. Big difference. Conclusion : The immunocompetence of SJZPS Vb 1 and SJZPS Vb 2 needs to be studied. It is worth investigating whether the similarities and differences in their structure and activity are related.
