临床上常用血管紧张素转化酶抑制剂(ACEI)治疗高血压。但ACEI有副作用,可引起咳嗽。目前对其发病机制还不清楚。有人认为ACEI活化花生四烯酸导致前列腺素样物质合成增加是导致咳嗽的原因。然而,环氧化酶作用于花生四烯酸产生的代谢产物不仅是前列腺素,还有前列环素和血栓素A_2。作者用抑制血栓素合成和拮抗血栓素/PGH_2受体且对环氧化酶活性没有作用的抗血小板凝集剂Picotamide,来评价ACEI所致咳嗽是否伴随血栓素合成的增加以及干扰血栓素途径是否能治疗咳嗽。
Clinically used angiotensin converting enzyme inhibitor (ACEI) treatment of hypertension. But ACEI has side effects, can cause coughing. The current pathogenesis is unclear. Some people think that ACEI activation of arachidonic acid lead to increased synthesis of prostaglandin-like substances is the cause of cough. However, the metabolites produced by cyclo-oxygenase on arachidonic acid are not only prostaglandins, but also prostacyclin and thromboxane A_2. The authors evaluated whether ACE-induced cough is associated with an increase in thromboxane synthesis and whether the thromboxane pathway is disturbed by using an antiplatelet agent Picotamide that inhibits thromboxane synthesis and antagonizes thromboxane / PGH2 receptors and has no effect on cyclooxygenase activity Cough treatment.
