[目的]利用重组汉坦病毒核蛋白及其单克隆抗体的酶标记物建立IgM捕获ELISA诊断试剂。[方法]制备、纯化重组核蛋白抗原和抗核蛋白单抗并进行辣根过氧化物酶标记,在抗人μ链包被板中结合四甲基联苯胺-过氧化氢(TMB/H2O2)酶底物系统,从血清中检测肾综合征出血热早期特异性IgM抗体,并与免疫荧光抗体法(IFA)进行比较。[结果]检测用经典免疫荧光抗体法确认的42份阳性血清和42份阴性血清,结果2种试剂检测结果差异无统计学意义。[结论]用重组汉坦病毒核蛋白及其单克隆抗体的酶标记物建立IgM捕获ELISA诊断试剂可用于肾综合征出血热的早期IgM抗体检测。
[Objective] To establish an IgM capture ELISA diagnostic kit by using enzyme markers of recombinant Hantavirus nucleoprotein and its monoclonal antibody. [Method] Recombinant nucleoprotein and antinuclear monoclonal antibody were prepared, purified and labeled with horseradish peroxidase (HRP). Tetramethylbenzidine - hydrogen peroxide (TMB / H2O2) Enzyme substrate system to detect early specific IgM antibodies to hemorrhagic fever with renal syndrome from the serum and to compare them with the immunofluorescence antibody method (IFA). [Results] There were 42 positive serums and 42 negative serums confirmed by classical immunofluorescence antibody method. There was no significant difference between the two reagents in the detection results. [Conclusion] The IgM capture ELISA diagnostic reagent can be used to detect early IgM antibody of hemorrhagic fever with renal syndrome by using enzyme markers of recombinant Hantavirus nucleoprotein and its monoclonal antibody.
