目的 :寻找与人肝再生增强因子 (hALR)相互作用的蛋白质 ,探讨其在肝再生过程中的分子生物学机制。方法 :采用酵母双杂交系统 ,以hALR作诱饵蛋白筛选预转化的人肝cDNA文库 ,对阳性克隆进行生物信息学分析。结果 :筛选出 6组与hALR具有特异性相互作用的蛋白基因 ,分别是 :血清白蛋白、金属硫蛋白、Na/K ATPase、硒蛋白P和两个未知功能基因的cDNA序列。结论 :初步克隆了与hALR相互作用蛋白基因 ,为以后深入研究这些蛋白质与hALR之间的相互作用 ,进一步揭示hALR的作用机制奠定了基础
OBJECTIVE: To search for proteins that interact with human augmenter of liver regeneration (hALR) and to explore their molecular biological mechanism during liver regeneration. Methods: The yeast two-hybrid system was used to screen pre-transformed human liver cDNA library with hALR as bait protein. The positive clones were subjected to bioinformatics analysis. RESULTS: Six protein genes were screened for specific interaction with hALR, including the cDNA sequences of serum albumin, metallothionein, Na / K ATPase, selenoprotein P and two unknown functional genes. CONCLUSION: The gene of protein interacting with hALR was preliminarily cloned, which laid the foundation for further study on the interaction between these proteins and hALR and further revealing the mechanism of action of hALR
